Potential for the Use of Polymerase Chain Reaction (PCR) in the Detection and Identification of Mycobacterium tuberculosis Complex in Sputum Samples

چکیده
The COBAS AMPLICOR Mycobacterium tuberculosis assay was evaluated for routine detection and identification of Mycobacterium tuberculosis complex (MTBC) in 629 sputum samples. In addition to the Polymerase Chain Reaction (PCR), diagnostic culture, which is considered the reference method, was also performed on theses samples. Of the 629 specimens tested, 64 were culture positive for M. tuberculosis and 51 were positive by COBAS AMPLICOR MTB assay and 19 specimens were considered PCR inhibitory. There were no false positive results in all tested samples and 3 PCR-negative, culture-positive specimens were considered false negative especially after reviewing the medical records for the 3 patients. A PCR inhibition rate of 3 % (19/629) was observed; these samples remained inhibitory despite re-testing. After excluding the inhibitory samples from the statistical analysis, the overall sensitivity, specificity, positive predictive value (PPV) and negative predictive values (NPV) for the COBAS AMPLICOR MTB assay were 94.4, 100, 100 and 99.5 % respectively compared to those of the diagnostic culture. In conclusion, the COBAS AMPLICOR PCR system proved itself a very useful component of today mycobacteriology laboratory.

RAPID DETECTION OF MYCOBACTERIUM TUBERCULOSIS IN CLINICAL SPECIMENS BY POLYMERASE CHAIN REACTION
We investigated the use of DNA amplification by polymerase chain reaction (peR) for detection of Mycobacterium tuberculosis in 300 patients who were suspected of having pulmonary tuberculosis and compared the results with culture results which were performed in parallel with PCR. Two-thirds of each sample was processed for smear and culture by standard methods and one-third was prepared fo...
متن کاملTHE DETECTION OF Mycobacterium tuberculosis BY PCR IN SPUTUM SAMPLES FROM LONG-TERMTREATED LEPROSY PATIENTS: ASSOCIATIONS WITH SKIN TEST RESULTS AND IMMUNOTHERAPY WITH Mycobacterium vaccae
As part of a series of investigations at Baba Baghi Leprosarium in lran, 44 long-treated leprosy patients were selected for our study. Samples of early morning sputum were obtained from each patient, examined by microscopy for acid-fast bacilli (AFB), and cultured for tubercle bacilli. These tests were negative, but the polymerase chain reaction (PCR) for an insertion sequence believed to b...
متن کاملEvaluation of in-house polymerase chain reaction assay sensitivity, can it be utilized in limited-resources settings?
Background :Polymerase chain reaction (PCR) assay has widely used for the detection of tuberculosis (TB). This study tried to compare in-house PCR with some well-known commercial PCR kits for detection of TB agent. Methods : Clinical samples obtained from 620 TB suspected patients were analyzed for the diagnosis of Mycobacterium tuberculosis complex (MTC) by in-house PCR. All samples were o...

Background: Molecular detection has recently been proposed by nucleic acid amplification, known as polymerase chain reaction (PCR). The aim of this study was to compare the diagnostic method of smear and polymerase chain reaction with culture in terms of sensitivity, specificity, positive and negative predictive value in the diagnosis of pulmonary tuberculosis. Methods: In this cross-sectional...
